Abstract

For a sperm DNA test to be clinically useful, it must have strong predictive capacity for pregnancy with little overlap between fertile and infertile samples. The four common tests used today are the sperm chromatin structure assay, the terminal transferase dUTP nick end labeling (TUNEL) assay, the sperm chromatin dispersion or halo test and the comet assay. Sperm DNA tests are all different and measure different aspects of DNA damage and have different sensitivities. Other topics discussed in the chapter are problems with DNA fragmentation test and why it should not be done, limitations and benefits of sperm DNA testing, and current concept about DNA test.
