Abstract

This chapter describes pharmacokinetics and metabolism. This chapter covers the absorption, bioavailability, distribution, half-life, metabolism, excretion, metabolites, neurosteroid metabolites, and specific metabolism. Progestins are rapidly absorbed with a longer half-life than progesterone and maintain stable levels in the blood. Progesterone is rapidly absorbed from the gastrointestinal tract and peak plasma concentrations are achieved within 2-3 hours. Progestogens are extensively bound to plasma proteins. Progesterone is metabolized primarily in the liver by reduction. It is reduced to a variety of active and inactive metabolites, including pregnanediol, pregnanetriol, and pregnanolone (pregnanedione).
