Abstract

Chromatography was introduced by Tswett in 1906 for separation of plant pigments. Chromatography is a laboratory technique used for the separation of closely related substances of macromolecules like proteins, amino acids, lipids and so on. Similar substances are separated by continuous distribution and redistribution between stationary phase and mobile phase. Separation depends on difference of size, and adsorption properties. Chromatography is set of techniques by which the member of a group of similar substances are separated by a continuous redistribution between two phases, one being mobile and the other stationary phase. It is used for qualitative screening, preparation, identification and quantitative analysis of wide variety of chemical compounds.
