This chapter aims to present a brief discussion on development of müllerian duct, clinical presentation, use of peritoneum in neovaginal lining, research hypothesis, and immunohistochemistry. The development from the müllerian duct is one of the most ill-understood topics in gynecology and remains an enigma. The fusion of the two müllerian ducts with subsequent canalization and hypertrophy leads to the development of adult female genital tract. Many surgical techniques like use of colon, intestine, and skin have been described for the creation of neovagina; however, none of them are successful in developing normal vagina. Immunohistochemistry tests (IHC) were performed on peritoneal biopsy samples and the presence of progenitor cells was demonstrated by OCT4 and SOX2 as markers. The immunostained images have been overlaid with the differential interference contrast image to show the localization of OCT4.
